STRASBOURG, France and CAMBRIDGE Mass., Nov 10, 2003 (Canada NewsWire via
COMTEX) -- Aventis and Vertex Pharmaceuticals Incorporated (Nasdaq: VRTX) today
announced voluntary discontinuation of phase IIb clinical trials of pralnacasan,
an oral interleukin-1 beta converting enzyme (ICE) inhibitor, in rheumatoid
arthritis (RA). This decision is based on results from an animal toxicology
study that showed liver abnormalities after a nine-month exposure to pralnacasan
at high doses. There have been no significant adverse events associated with
liver toxicity in subjects and patients who participated in pralnacasan studies
to date.

The decision was confirmed following a discussion with the Food and Drug
Administration (FDA) that Aventis and Vertex had requested. During this
discussion, the FDA supported the decision of Aventis and Vertex to discontinue
present phase IIb clinical trials in rheumatoid arthritis.
Also during this discussion, it was determined that two, shorter-term on- going
phase I trials will continue as planned, as the FDA agreed with Aventis and
Vertex that the toxicity findings based on longer-term regimens in animals did
not imply safety concerns in the significantly shorter phase I trials.
"This is an unexpected toxicology finding, and the prudent action is to
discontinue the ongoing phase II clinical studies, and fully evaluate the
toxicology results before moving ahead," said Frank Douglas, Executive Vice
President of Drug Innovation & Approval and Member of the Aventis Management
Board. "We are committed to working with Vertex to better understand and
hopefully resolve this issue."

of investigational therapeutic agents in clinical trials and ultimately,
clinical practice. Today`s decision is based on the results from a 9-month
animal toxicology study.
The findings observed in the 9-month toxicology study were not observed in prior
toxicology studies conducted with pralnacasan, including 6-month studies in two
different species. Decisions on further clinical trials of pralnacasan will be
made after the final analysis of the nine-month toxicology study and data from
an on-going 12-month toxicology study are available.
Phase IIb Rheumatoid Arthritis Clinical Trials

randomized, placebo-controlled, double-blind, multi-center clinical trial to
evaluate the safety and efficacy of pralnacasan in patients with RA. During this
study, patients were permitted to continue background methotrexate therapy, the
industry standard treatment for rheumatoid arthritis, which also can induce
liver toxicity. Patients completing this trial were allowed to roll over to an
extension trial where they were treated with pralnacasan or standard of care
therapy. The discontinued phase IIb studies were to evaluate 340 patients
treated with pralnacasan or placebo for 12 weeks. Approximately 330 patients had
been randomized prior to the decision to discontinue the study, and
approximately 50 had completed treatment.

BOSTON (CBS.MW) -- GlaxoSmithKline and Vertex Pharmaceuticals won U.S. clearance for an AIDS-fighting drug that the companies say will offer more convenient dosing for patients.
Shares of Vertex (VRTX: news, chart, profile), a Cambridge, Mass.-based biotech firm, gained 54 cents, or 4.1 percent, to $13.57. U.S.-listed shares of GlaxoSmithKline (GSK: news, chart, profile), a U.K.-based pharmaceutical giant, edged up 39 cents to $43.92 after earlier touching a new 52-week high of $44.03.
The Food and Drug Administration approval, announced by the companies Tuesday morning, had been expected.
GlaxoSmithKline and Vertex said their new medication is the only one in its category -- protease inhibition -- that offers "flexible dosing with no food and water restrictions."
The drug, called Lexiva, can be taken once or twice daily. Patients with HIV, the virus that causes AIDS, typically take numerous medications throughout the day and often have difficulty following the complicated drug regimens. Drugs that are dosed less frequently and with fewer restrictions can ease the burden on patients.
Doug Manion, GlaxoSmithKline`s vice president of clinical development, said in a statement that the new drug illustrates an "ongoing commitment to providing flexible anti-HIV therapies for patients."
FDA approval of Lexiva is also important for GlaxoSmithKline`s business. The company has long been a leader in HIV medications, but it`s facing increased competition from drugs such as Gilead Sciences` (GILD: news, chart, profile) Viread.
The FDA OK marks a milestone for Vertex as well. Lexiva will become the biotech firm`s second drug on the market. The other, called Agenerase, is also an HIV medication that`s partnered with GlaxoSmithKline.
Merrill Lynch in a note to clients predicted combined sales of Agenerase and Lexiva would be $61 million this year and $100 million next year.

Es sind auf jedenfall Medikamente mit hohem Potenzial dabei,aber es dauert etwa 2-3 jahre bis die auf dem Markt kommen.
Die Aktie ist zu den aktuellen Kurs ein Kauf .
Dafür spricht der Cashbestand ,der fast genauso hoch ist wie die Marktcap.
Das bedeutet das man die Produkte praktisch kostenlos dazu bekommt.

(viele Produkte in der frühen Phase, wo sie mit einer großen Wahrscheinlichkeit scheitern. Doch ich sehe auch 2 Lichtblicke)

@Dausend: Also ich habe mit Biotechs in den letzten Monaten ganz gut verdient - ich bin überzeugt, daß dies die Branche (zumindest) dieses Jahrzehntes ist .
Vertex werde ich mir morgen mal ansehen, genauso Immunomedics - ich mag abgestürztr BioTechs besonders gern.....

Nö, aber die Aktie hatte trotzdem irgendwann im letzten Jahr 25 oder mehr % verloren und dann habe ich halt zugesnappt und gespielt (einmal ab 7.50 und einmal ab 5.25 USD). Vertex habe ich mir ein paar gekauft, Cash > 8 USD.

ein paar Überlegungen von mir zu Vertex:
1.) Ich denke, über die Produktpipeline genau diskutieren zu wollen, macht für medizinische Laien keinen großen Sinn. Ich halte also nicht viel davon, wenn hier darüber geredet wird, dass Vertex potentielle Blockbuster in der Pipeline hat, oder eben nicht (oder ist ein erfahrener Mediziner unter uns - uns selbst die irren sich oft bezüglich Marktchancen von Medikamenten). Das ist generell das Problem von Biotechs, mit dem man leben muss.
2.) Was man nur dazu sagen kann ist, dass Vertex wohl kein Medikament in der 3. Phase hat - das nächste ist so gegen Ende der 2. Phase und somit noch einige Zeit von der eventuellen Einführung entfernt. Große Fantasie dürfte also im Moment nicht bestehen. Und auch das aktuell auf den Markt gebrachte Medikament wird nicht als umsatzstark eingeschätzt. Auf den 1. Blick sieht es also mau aus, ABER: das gute daran ist, dass das alles bekannt ist. Man kann also davon ausgehen: eingepreist. Positive Überraschungen sind also drin. In etwa: Das neue Medikament verkauft sich doch besser als erwartet oder es gibt Fortschritte mit der Pipeline. Damit rechnet im Moment niemand. Das ist kurzfristig die Chance.

Ach, noch eine Sache wegen dem Cashbestand: Der ist in der Tat fast so hoch, wie der aktuelle Wert von Vertex, aber:
- Die Firma verbrennt im Moment noch ordentlich Geld (etwa $ 150 Mio. pro Jahr) und das ändert sich auch nicht, wenn nicht neue Medikamente kommen oder sich eingeführte besser verkaufen - ein ganz schönes Risiko. Man kann also nicht sagen, man bekommt die Pipeline geschenkt - das ist nur eine Momentaufnahme.
- Was interessant wäre zu wissen: kosto1929 schrieb mehrere Male in diesem Thread: "Vergesst die Schulden nicht." Das würde mich interessieren. Wer weiß, wieviele Schulden Vertex aktuell hat? Ich konnte nur finden, wieviel Geld sie im Moment verbrennen. Wenn auch noch höhere Schulden dazu kommen, dann steigt das Risiko meiner Meinung nach schon etwas an bei so einem Cash-Burner Also: Wer weiß, wieviele Schulden die haben? @kosto1929: Du hast die Daten wohl auch aus Yahoo-Finance. Auf der Seite war ich auch. Was bedeutet die Kennzahl Total Debt? Sind das sie Schulden? Das wäre ja Wahnsinn. Dann sollte man sich das doch nochmals überlegen. Das würde das Cashpolster in einem ganz anderen Licht erscheinen lassen . Wenn nicht - woher hast Du die Infos, die Dich zu der Aussage: "Vergesst die Schulden nicht!" führen?

Das ist zur Zeit Fakt. Freilich können da gute News kommen, aber warum dann jetzt schon einsteigen? Es gibt zig bessere Bios, die Phase III Produkte haben und kurz vor der Gewinnschwelle stehen.
Fazit: Für Zocker ganz lustig, für Langfristinvestoren gilt, entweder Finger weg oder wenn man auf Verlusten hockt, abwarten und bei besseren Fundamentals nachkaufen. Die letzten Zahlen waren jedenfalls Horror (auch ohne die Medikamenteneinstellung)

Sehe ich auch so. Habe selber Napro, Millennium und Geron im Depot. Also HumanGenome oder Vertex würde ich nicht unbedingt kaufen. CV Therapeutics hab ich leider noch, aber da ging ja so einiges schief. Schade eigentlich.
Bei Napro kann man wirklich nicht von Überbewertung sprechen. Mille hat einen hohen Cash-Anteil und gibt eben viel Geld für die Forschung aus.
Die Kursziele der Kulmbacher waren schon immer etwas daneben... Rambus 500 $ z.B.... nunja wenn sie damals den Rechtstreit gewonnen hätten... Ich sehe das alles recht positiv. Mille hab ich bei 7,5 € fleißig nachgekauft, hat sich gelohnt auch wenn ich noch bei 10% im Minus liege. Sollte Mille ohne Grund 30-40% nachgeben kaufe ich eben wieder nach... in ein paar Jahren steh ich dann sicher auch bei 50-100% plus und schlage trotz meines Erstkaufs bei 99 € (ja ich weiß war so ziemlich die Spitze) jdes Sparbuch

CAMBRIDGE, Mass., Feb 23, 2004 /PRNewswire-FirstCall via COMTEX/ -- Today in Nature Medicine, researchers from Vertex Pharmaceuticals Incorporated ( VRTX ) have demonstrated for the first time that a selective small molecule inhibitor of the Aurora kinases, VX-680, profoundly reduces tumor growth in cancer models. Aurora kinases are known to be overexpressed in many tumor types, including colon cancer, breast cancer, and leukemia.
Research into the function and activity of Aurora kinases over the past several years has suggested that they may play multiple roles in the development and progression of cancer, by acting as regulators of cell proliferation, by transforming normal cells into cancer cells, and by downregulating p53, one of the body`s natural tumor suppressors.
In the study reported today, the small molecule Aurora kinase inhibitor VX-680 blocked cancer cell proliferation, and also triggered cell death in a broad range of tumor cell types. Data from in vivo xenograft models indicated that VX-680 achieved complete inhibition of tumor growth at well-tolerated doses, and in some instances, tumor regression was observed. The report suggests that Aurora kinase inhibition provides a promising, novel approach for the treatment of multiple human malignancies.
"Aurora kinases represent a potentially important class of targets for the future treatment of cancer, and we have now demonstrated for the first time that a small molecule Aurora kinase inhibitor not only blocks tumor cell proliferation but also induces tumor cell death," commented Karen Miller, Ph.D., Director of Biology, Vertex Europe and senior author of the study. "The ability of VX-680 to cause tumor regression is particularly exciting."

Aurora Kinases and Cancer
Aurora kinases (also known as BTAK and STK15) are a family of serine- threonine kinases that have been strongly linked to tumorigenesis. Aurora kinases, which play a central role in controlling cell division, are disregulated in many types of human cancer, including leukemia, colon and breast cancer. Overexpression of Aurora kinase has been shown to promote the transformation of normal cells into cancer cells, and decreased Aurora kinase activity is associated with enhanced function of the body`s normal tumor suppressor genes. Amplification of Aurora genes is associated with progression of colorectal cancer and poor prognosis in certain types of breast cancer.
Cancer is the second leading cause of death in the United States; more than 1 million solid tumor cancers are diagnosed in the United States annually, with approximately 500,000 deaths. More than 200,000 new cases of breast cancer and 150,000 cases of colorectal cancer are diagnosed in the U.S. each year. The five-year relative survival rates for patients with metastatic breast cancer and patients with colorectal cancer are 21% and 8% respectively. There are more than 30,000 new cases of leukemia in the U.S. every year, and more than 20,000 deaths.
Cancer cells typically contain mutations in a number of genes, which ultimately results in uncontrolled cell growth and tumor metastasis. As enzymes specific for and essential to cell growth and division, Aurora kinases hold the potential to be important control points for slowing the growth and spread of tumors.
About VX-680
VX-680 was discovered at Vertex as part of a broad research effort targeting the kinase gene family. Vertex researchers published the three- dimensional atomic crystal structure of Aurora-A kinase in 2002, a key scientific advance that enabled the design and optimization of multiple classes of small molecule Aurora kinase inhibitors. VX-680 was advanced to preclinical development in 2002, following evaluation of the compound`s activity in cell lines and in animal models of tumor growth. In the study published today, Vertex researchers reported that VX-680 induced tumor regression of 22% in a human pancreatic xenograft model and of 56% in a human colon cancer xenograft model. Vertex has filed an investigational new drug (IND) application with the FDA to support Phase I clinical development of VX- 680 in 2004.
Vertex`s broad kinase research program is supported by a collaboration with Novartis Pharma AG begun in May 2000 and amended in 2004. Under the terms of the amended agreement, Vertex may opt to continue development of VX- 680 under the terms of the original agreement, or to develop and commercialize VX-680 independently.
The research was published in the March 2004 issue of the scientific journal Nature Medicine. Cancer cells typically contain mutations in a number of genes, which ultimately results in uncontrolled cell growth and tumor metastasis. As enzymes specific for and essential to cell growth and division, Aurora kinases hold the potential to be important control points for slowing the growth and spread of tumors.
About VX-680
VX-680 was discovered at Vertex as part of a broad research effort targeting the kinase gene family. Vertex researchers published the three- dimensional atomic crystal structure of Aurora-A kinase in 2002, a key scientific advance that enabled the design and optimization of multiple classes of small molecule Aurora kinase inhibitors. VX-680 was advanced to preclinical development in 2002, following evaluation of the compound`s activity in cell lines and in animal models of tumor growth. In the study published today, Vertex researchers reported that VX-680 induced tumor regression of 22% in a human pancreatic xenograft model and of 56% in a human colon cancer xenograft model. Vertex has filed an investigational new drug (IND) application with the FDA to support Phase I clinical development of VX- 680 in 2004.
Vertex`s broad kinase research program is supported by a collaboration with Novartis Pharma AG begun in May 2000 and amended in 2004. Under the terms of the amended agreement, Vertex may opt to continue development of VX- 680 under the terms of the original agreement, or to develop and commercialize VX-680 independently.
The research was published in the March 2004 issue of the scientific journal Nature Medicine. The authors of the paper, titled "VX-680, a Novel Small Molecule Inhibitor of the Aurora Kinases, Suppresses Tumor Growth In Vivo," are Elizabeth A. Harrington, David Bebbington, Jeff Moore, Richele K. Rasmussen, Abi O. Ajose-Adeogun, Tomoko Nakayama, Joanne A. Graham, Thierry Hercend, Anita Diu-Hercend, Michael Su, Julian M.C. Golec, and Karen M. Miller, all current or former employees of Vertex, and Cecile Demur of INSERM.

Das amerikanische Biotech-Unternehmen Vertex Pharmaceuticals Inc., ein Spezialist für Wirkstoffe gegen Viruserkrankungen, und die Basler Novartis AG, Europas drittgrößter Pharmakonzern, nahmen eine Änderung an ihrem Kooperationsabkommen vor.
Die Partner arbeiten bei der Entdeckung und Entwicklung von Medikamenten zusammen. Nun bleibt Vertex verantwortlich für die Entdeckung von Arzneimittelkandidaten, aber Novartis (Virt-X: NOVN.VX - Nachrichten) Anzeige
kann unabhängig von Vertex eigene Forschungen durchführen. Zudem ist Novartis verantwortlich für die pre-klinische und die klinische Entwicklung.
Gestern stiegen die Aktien von Vertex an der NASDAQ (NASDAQ: Nachrichten) um 0,1 Prozent und verließen den Handel mit 9,80 Dollar.
